Not according to society. We don't perform a chromosome check to determine whether to label people male or female.
If the Y chromosome is your barometer, then a male gave birth. The NCBI declares this person a female, despite the presence of a Y chromosome. This isn't the only time this has happened either.
Society, nor the NCBI, can dictate and or change immutable facts.

These are psychological/sociological questions and are highly dynamic. People who will be whoever their nature drives them to be, and if their nature drives them to falsely label themselves something they are not, then that is always subject to change (non-immutable).

They don't check genotype because it costs money to do so. People don't wanna pay to check that when they're paying for a bunch of other tests to make sure the baby is simply okay and healthy.

I personally think people should just accept who they are...and stop trying to be something they aren't or never will be.
Trans people do exactly that, but it seems that some in society want people to be what they want them to be something they aren't - their genitals.
To put it another way, I tried to be (and wanted to be) a guy for most of my life. I finally gave in and admitted that it wasn't me and that I was just trying to be what society wanted me to be. I've finally accepted who I am. However, some folks are of the belief that the only thing that matters in determining my identity, for all intents and purposes, is what the doctor saw when I was born. Go figure.
