We cannot synthesise it due to a mutation in the GULO gene. Normally GULO is an enzyme that catalyses the reaction of D-glucuronolactone with oxygen to L-xylo-hex-3-gulonolactone. Our non human primate ancestors lost the ability to do so about 61 million years ago.
