I agree totally with drfeelgood. In MY world, that misunderstanding would mean that she wasn't thinking like you - she's not thinking about going out with YOU the way you are with HER.
However, with such a close friend, sometimes this is something she NEEDS to consider. Recently, I started dating my best friend - which I put off for a VERY long time because I was afraid of what might happen to our relationship (along with life's usual complications - but I THINK that was the big thing). I'd bet that she really loves you (love != dating) and she simply thrives on the relationship you have. There is a good deal of miscommunication in these relationships as girls and guys think differently - and she might be very content with where you are now.
On the other hand, I would really talk to her honestly, because at some point she needs to understand (for herself at least) that an exclusive, close relationship can't last in simple friendship. Your intimacy can (not sex - closeness) but an exclusive relationship where she is also YOUR best friend doesn't always work if you date a different girl.
Of course there are exceptions - I don't have enough information as feelgood said. You don't lose friends (or shouldn't) when they date other people. Just as in MY case at least, I had a dating relationship with my best friend without actually dating her.
