Ooh, good question! I'll have to give it some thought. It depends heavily on your definition of "significant" though. I'd say Sayid is probably a good possibility at this point, but I'm not sure they're going to kill someone off.

I don't think anyone will die before the break -- only because someone on the verge of death would make a cliffhanger that would drive us all nuts for 13 weeks.
