Booting into Safe Mode usually allows you to manipulate your system in ways you cant when regularly logged in...hit F8 repeatedly till the screen pops up that allows you to choose.

Warning: If you dont know what youre doing you can really damage your computer while in this mode...

Most files/programs can be located in:

C:\Program Files\

and like tekwerx said, if you can't delete them regularly, you can in safe mode. But be careful in safe mode cause you can really mess things up.

I've ran into this a couple of times... if nothing else works, sometimes you can get small programs off the developers website or just anywhere online (that's risky though, watch out for viruses, trojans, spyware, etc.) that will act as the uninstall file and automatically do the job.

Assuming you're running Windows, if you go to:

Start --> Control Panel (hunt around through the submenus if you're not running XP) --> Add or Remove Programs

You'll be presented with a list of installed programs, and it'll let you uninstall the program. Try that first. :)

Any program files (besides spyware which is running in the background of course) can be uninstalled by navigating to C:\Program Files in your "My Computer" tool. Just right click on the folder and select "Delete".

If the process is still running in the background it gets trickier. Then you have to reboot in Safe Mode to do it or stop the process from running in Task Manager. You can also boot into Safe Mode with Command Prompt and manually type your way to the directory.

My favorite way of doing it is either with the Command Prompt or using the Recovery Console.

This is good advice. Be careful deleting folders. Let Windows do it.
