They revealed that some mysteries will be resolved by the end of the season: what was up with the guys in the arctic, Walt's ability to rejuvenate animals, the "magic box" idea. They also said that Patchy's resurrection will be explained within the next couple of weeks. The smoke monster will not be explained until the end of the series.

They will be revisiting the psychic's predictions for Claire in the future as well as Walt's abilities. The Others' obsession with children and pregnancy goes beyond the inability to successfully bear children on the island. The Other's definitely have an interest in children with special abilities. That's why they took Walt.
For the (insert insanely large number here) time, Christian Shepherd is DEAD! We have not seen Jacob in a previous episode and won't until he is revealed. There will be a Rousseau flashback eventually. In it, it will talk about Montand and his arm. Source: Lost Podcasts

Rousseau has to overcome her issues before she can approach her daughter. "The Others are a lot badder than you think they are." By the end of the season you may not think that their act of hanging Charlie was out of character for them. This seems to imply/confirm that Ethan/The Others are in fact responsible for Charlie's hanging. Source: Lost Podcasts
Cuse and Lindelof revealed: We have a very clear plan for this season, and I don't think we'll get back to Michael and Walt's story this season. Harold Perrineau's story is not finished. He is not on the show currently, but I think everybody is very curious to know what happened to Michael and Walt, and we hope to get back to that story. That character is still out there in the Lost universe. [Eko might be back and it] might not necessarily be a flashback episode.
