Well, the one that popped into my mind isn't a note, but a wolf howling. It's the beginning to 'Dig up her Bones' by the Misfits.

Me, my friend Rob and a couple of girls were at a concert of a heavy punk band called 'Loco' out of Winnipeg. For their last song, the sound of a wolf howling wailed over the speakers. We knew what was coming, so like a couple of misfits; into the mosh pit we went!

This is an easy one for me.

The Eb in "Hell's Kitchen" by Dream Theater. John Petrucci plays this brilliant solo, and then it goes into a great climatic few chords, which starts with Eb. The whole reason I listen to the song is that few seconds of music.

Notable mention also goes to Liquid Tension Experiment - Biaxident Towards the end of the song, John Petrucci has another brilliant moment and plays a melody rather high on the neck with the neck pickup. That melody is dripping with emotion. It's great.
