I haven't much time, but I wanted to mention that Matt Ridley's Nature via Nurture (thank you, Barnes & Noble clearance aisle) has some nice information re: the title of this thread, with footnotes and all.
The gist is that one or more hox gene(s) can superficially account for the developmental differences between chimpanzee and human brains. It is, naturally, somewhat more complicated, but the suggested impossibility of positive mutations for brain development is seemingly empty.
